Method for producing L-2-aminobutyric acid by fermentation induced by IPTG

L-2-aminobutyric acid is a non-natural hand-type amino acid that inhibits the transmission of human neural information, enhances the activity of glucose phosphatase, and promotes brain cell metabolism. Currently, the synthesis of L-2-aminobutyric acid includes chemical methods and biological methods. The chemical method involves removing the methylthio group from L-methionine under the action of Raney Ni to form L-2-aminobutyric acid and using a chiral resolving agent to form a diastereomer with racemic 2-aminobutyric acid. Resolution gave L-2-aminobutyric acid. Biosynthetic methods include enzymatic methods, fermentation methods, and so on.
Whether it is organic synthesis or chemical resolution in chemical methods, the cost is relatively high. At present, the biosynthesis of L-2-aminobutyric acid has the advantages of mild reaction conditions, high process efficiency and good stereoselectivity, and has a broad industrial application prospect. A method for producing L-2-aminobutyric acid by induction of fermentation by IPTG is described below [1]. Proceed as follows:
(1) The strain is connected to the seed tank, and the culture is carried out under the conditions of a temperature of 33°C to 37°C, pH 6.5~7.5, dissolved oxygen of 15% or more, and OD 9-25. Mature strains.
(2) The mature strain is inserted into the fermenter and cultured under the following conditions: through deep liquid ventilation fermentation, pre-fermentation (before 15h), the IPTG with a final concentration of 0.1-0.3 mmol/L is used in one time through microfiltration filtration sterilization technology, or the required IPTG is added before 15 hours by the fed-batch technique, the temperature of the fermentation process is controlled in two steps, and the temperature in the early stage of fermentation is controlled at 34-37°C. When the cells are grown to the mid-log phase, the temperature is adjusted to 30-33°C for culture.
(3) After the fermentation, the fermentation broth is first sterilized by ceramic membrane, and then passed through an ultrafiltration membrane of 2-4Kd molecular weight, decolorized by activated carbon, adsorbed by 732 type ion exchange resin and eluted with ammonia water, and concentrated by evaporation to obtain 2-aminobutyric acid.
The purity of the product produced by the above fermentation method can reach more than 95%, which greatly improves the production capacity and efficiency of L-2-aminobutyric acid, and can satisfy the large-scale industrial production of L-2-aminobutyric acid.
