Composition and storage of bovine serum albumin

2019-07-26 来源:亚科官网
Bovine serum albumin(BSA) is widely used in biochemical experiments which plays a very important role. It is used as a stabilizer for limiting activity of enzymes or modifying enzymes in preservation solutions and reaction solutions, and preventing decomposition and non-specific adsorption of enzymes.

Composition of BSA
Although the large part of serum composition is known, there are still to be solved. The known components in BSA are probably as follows.

1. Protein is the main component in bovine serum. In addition to carrying metal ions, fatty acids and hormones, it is mainly albumin, globulin. Fibronectin cells promote cell adhesion; α2 macroglobulin inhibits trypsin; fetal bovine serum contains fetuin to promote cell attachment; transferrin binds to iron ions, reducing its toxicity and being utilized by cells.

2. Polypeptide. Platelet growth factor can promote cell division which is one of the major members of the polypeptide family. It is the main cell proliferation factor. Although the content of Fibroblast growth factor, epidermal growth factor, nerve cell growth factor and so on in serum is rare, it also has an effect on cell growth.

3. Hormone and Insulin promotes the cell’s uptake of glucose and amino acids, which is involved in cell division. Insulin-like growth factor binds to the insulin receptor expressed by the cell, and thus has the same effect as insulin. Growth hormone promotes cell proliferation.

4. Other ingredients include amino acids, glucose, ketoacids etc. Trace elements bound to proteins are of interest for cell culture.

Storage of BSA
Dissolve 10 grams BSA with 100 of water, or dissolve in PBS. Then pack it into small sticks. If you do not use preservatives, you can store them in a thermostat at minus 20 or 30 degrees Celsius. If preservatives are used, they can be stored at a temperature of 4 degrees Celsius. They can be stored for several months without deterioration. Preservatives may affect the effect of BSA and should be used with caution.
