The nature and development of 3,3',5,5'-tetramethylbenzidine (TMB)

3,3',5,5'-Tetramethylbenzidine (CAS: 54827-17-7) is a new type of safe chromogen reagent, generally referred to as TMB, which is a white or light yellow crystalline powder. The item No. of Yacoo is S0025, the molecular formula is C16H20N2, the molecular weight is 240.34, and the melting point is 168-171°C. It is odorless, tasteless, hardly soluble in water, and easily soluble in organic solvents such as acetone, ether, dimethyl sulfoxide and dimethylformamide. Compared with traditional chromogen reagents, TMB has the advantages of high detection sensitivity, low toxicity, safety and reliability, and replaces the traditional carcinogen, benzidine. Since the amino group is heated and unstable, it should be stored at 2-8°C, dry and protected from light.
As for the synthesis and application of TMB, as early as 1974, Holland et al. synthesized 3,5,3,5'-tetramethylbenzidine, and use it for the blood test, Holland et al. also continued subcutaneous injection of benzidine and TMB in rats, the results showed that benzidine has carcinogenic effects, while TMB does not induce tumors. In addition, Garner and other tests have proved that TMB has no mutagenic effect. Based on this, we first synthesized TMB and its hydrochloride and sulfate in China and successfully applied it to blood mark inspection.
At present, TMB has gradually replaced the strong carcinogen benzidine and other carcinogenic benzidine derivatives for clinical testing, forensic testing, criminal detection and environmental testing. Clinically used for the diagnosis of hepatitis, the results within 30 minutes, rapid and accurate; forensic examination of blood marks, the discovery of occult blood fingerprints; environmental aspects for the detection of residual chlorine in water. In recent years, in clinical biochemical tests, 3,3',5,5'-tetramethylbenzidine as a new substrate for peroxidase, in enzyme immunoassay (EIA) and enzyme-linked immunosorbent assay (ELISA) ) has been widely used.
As a chromogenic reagent, TMB has a relatively high stability and low toxicity, making it more widely used than the traditional carcinogen, benzidine. Its own reducibility and its ability to participate in the reaction also make the reagent more and more widely used. Its derivatives TMB hydrochloride, sulfate, propane sulfonate, etc. are also used in the application of TMB. It was synthesized and applied in the study.
However, due to the late start of our domestic chemical industry, the CAS number of TMB series products is commonly used in China. For example, TMB sulfate, TMB propanesulfonic acid, TMB propanesulfonate, TMB sulfate CAS number: 54827-18-8, TMB propanesulfonic acid CAS number: 102062-36-2, TMB sodium propane sulfonate CAS number: 10262-46-4, and the CAS number of these things in our country is basically 102062-36-2, which is the CAS number of TMB propanesulfonic acid. Foreign databases are clearly listed, but domestic is difficult to distinguish.
