Application of MOPS|CAS:1132-61-2| in Cholesterol Detection Kit

2022-04-15 来源:亚科官网

MOPS is a zwitterionic buffer discovered by Good et al. in the 1960s. It has a pKa value of 7.14 at 25°C and an effective pH adjustment range of 6.5-7.9. It is commonly used in detection kits, cell culture media and other fields. This week, YACOO editors introduce the application of MOPS in cholesterol detection kits.

The detection kit in the CN109613282A consists of two parts: reagent R1 and reagent R2.

The reagent R1 consists of 5 parts, specifically: surfactant (one or more of ceteth-23, Tween-60 and Span-60, 10.0-30.0mmol/L), magnesium chloride (5.0 -15.0mmol/L), 4-aminoantipyrine(0.2-2.0mmol/L), MOPS buffer with pH 7.0 (50.0-150.0mmol/L) and tert-butyl-4-hydroxyanisole ( 0.1-0.7g/L);

Reagent R2 consists of 7 parts, specifically: cholesterol esterase (0.2-3.0KU/L), cholesterol oxidase (1.0-5.0KU/L), peroxidase (5.0-15.0KU/L), deoxycholic acid Sodium (40.0-70.0mmol/L), developer (2,4,6-tribromo-3-hydroxybenzoic acid, sodium 2-hydroxy-3-m-toluidine propanesulfonate, phenol and 4-chlorophenol) One or more of 1.0-5.0mmol/L), magnesium chloride (5.0-15.0mmol/L) and MOPS buffer (50.0-150.0mmol/L) with a pH of 7.0.

The determination kit has simple components, and the accuracy is significantly improved by adding tert-butyl-4-hydroxyanisole; Using the selective inhibition method, the surfactant selected for reagent R1 exerts the effect of scavenging non-HDL-C at the optimal concentration; and the amount of the sample required in the present invention is small (3μL serum is sufficient), and it is basically not affected by hemoglobin, triglyceride, bilirubin, etc.;
