  • Purpose：
    Advanced liver cancer treatment ;Sorafenib(BAY 43-9006)是一种新型的，有效的RAF激酶和VEGFR双靶点抑制剂，可以抑制Raf-1 (IC50=6 nmol/L), 野生型BRAF (IC50=22 nmol/L)和V599E突变的BRAF (IC50=38 nmol/L), 以及 VEGFR-2 (IC50=90 nmol/L)和VEGFR-3 (IC50=20 nmol/L)，但是并不抑制MEK-1 or ERK-1 活性 (IC50 >10,000 nM)。Sorafenib具有广泛的抗肿瘤活性，在结肠癌，乳腺癌和非小细胞肺癌移植瘤模型中抑制肿瘤细胞增殖和肿瘤生长[1]。Sorafenib抑制MEK和ERK磷酸化，并下调PLC/PRF/5和HepG2细胞中细胞周期蛋白D1的表达。
