The advantages of Tris-HCl buffer
Alkaline is strong, it can be used to configure pH buffer from acid to base;

Shortcomings of Tris-HCl buffer
pH is greatly affected by concentration, diluted 10 times, The change of pH is greater than 0.1
Large temperature effect, such as pH=8.4 at 4°C and pH=7.4 at 37°C
Easy to absorb carbon dioxide in the air.
Some interference with some pH electrodes, electrode compatible with Tris solution should be used if necessary
In addition to Tris-HCl, tris also has a variety of derivatization buffers:
TBS=Tris-HCl+ NaCl+KCl, commonly used for washing immunostained tissues or Western blotting membranes in Western Blotting;
TBST=Tris-HCl+NaCl+tween20, a membrane buffer commonly used in Western Blotting;
TE=Tris-HCl+EDTA, which is protective against DNA bases and is often used for DNA stabilization and storage;
TAE=Tris base+acetic acid+EDTA, which is a buffer system widely used in short-segment DNA electrophoresis;
TBE=Trisbase+boric acid+EDTA is suitable for long-term DNA electrophoresis and has better separation effect on smaller fragments.
