Application of Triton X-100 for IVD

2020-06-16 来源:亚科官网

Triton X-100 is a non-ionic surfactant with high stability and good compatibility with other types of surfactants. It is used in chemical, textile, water treatment and other industrial fields. It is also used in life sciences and in vitro diagnostics.

Triton X-100 has a hydrophilic end and a hydrophobic end. On the one hand, it does not dissociate in water. It has high stability in solution, is not easily affected by strong electrolyte inorganic salts, and can be combined with phospholipids and other lipids in biofilms. It forms a soluble complex; on the other hand, the hydrophobic end can also combine with the hydrophobic region of the membrane protein to form a complex and dissolve in the solution. Therefore, Triton X-100 is often used to help separate proteases in water, for nucleic acid extraction and preparation of in vitro diagnostic reagents.

Triton X-100 is used for nucleic acid extraction: Triton X-100 can dissolve lipids to increase the permeability of cell membranes to antibodies [1]. In nucleic acid lysate, Triton X-100 can cooperate with SDS to improve the ability to cleave the surface membrane structure of cells or enveloped viruses, and promote the release of nucleic acids. In addition, residual Triton X-100 after rinsing During the molecular diagnosis process, such as PCR amplification, it can antagonize the residual SDS and suppress the impact of the residual SDS on the subsequent process [1].

Triton X-100 is used in a variety of in vitro diagnostic reagents, such as creatinine detection, blood lipid typing determination, cholesterol determination, and total bilirubin detection kits.
