β-nicotinamide mononucleotide(NMN) has important physiological functions for human cells and is closely related to human immunity and metabolism. Studies have shown that β-nicotinamide mononucleotide can delay aging and has special medical application value. People are committed to developing efficient synthetic methods to achieve large-scale production of β-nicotinamide mononucleotide.

The synthesis method of β-nicotinamide mononucleotide mainly includes two ways, namely biosynthesis and chemical synthesis.

In the biosynthesis of β-nicotinamide mononucleotide, nicotinamide and 5'-phosphate ribosyl-1'-pyrophosphate are used as substrates to catalyze the preparation of

β-nicotinamide mononuclear under the action of nicotinamide phosphoribosyl transferase Glucuronide. The price of enzyme in this method is higher and the source is limited. Therefore, when this method is used to prepare β-nicotinamide mononucleotide, the production capacity is low and the production cost is high. In recent years, through research and development and improvement, the yield of β-nicotinamide mononucleotide prepared by biosynthesis has been improved, and the cost has gradually decreased.

Wei Zhu and Xiaofei Zhang developed a Catalytic preparation of β-nicotinamide mononuclear by immobilized whole-cell one-step enzymatic reaction,the synthetic amount and conversion rate of β-nicotinamide mononuclear enzymatic reaction can be increased, the immobilized whole-cell can be used repeatedly, and the reaction complexity and production cost can be reduced. According to the method, D-5-ribose phosphate, ATP and nicotinamide are used as raw materials, beta-nicotinamide mononucleotide can be efficiently and biologically synthesized by catalysis of immobilized whole-cell containing phosphoribosylpyrophosphate synthetase and nicotinamide phosphoribosyltransferase.The synthetic route is as follows:

In chemical synthesis, a substance with a furanose structure is generally selected as a substrate, and a multi-step functional group conversion is performed to obtain a β-nicotinamide mononucleotide molecular skeleton, which is then eluted through a resin column to obtain a β-nicotinamide mononucleotide aqueous solution product. This method has a large selection of synthetic routes and a relatively wide range of raw material sources, but there are many steps and the yield is not high,and it is not conducive to subsequent purification.
