Application of nucleic acid dissociation solution added with guanidine isothiocyanate|CAS593-84-0| in nucleic acid extraction and purification

During the outbreak of the COVID-19, rapid nucleic acid diagnosis is extremely important for the prevention and control of the epidemic. Nucleic acid sample extraction is divided into two parts: protein and nucleic acid dissociation and nucleic acid extraction. The current nucleic acid extraction and purification mostly use resin adsorption or magnetic bead adsorption, which requires a high-speed centrifuge or automatic nucleic acid extraction instrument, which takes a long time. In order to solve the existing problems, Li Xiaofeng and others have developed a nucleic acid dissociation solution. The dissociation solution is composed of guanidine isothiocyanate, water-saturated phenol, alcohol reducing agent and 8-hydroxyquinoline. The components and their concentrations and pH values are as follows[1]:

Guanidine isothiocyanate: the molar concentration is 1mol/L-2mol/L;

Water saturated phenol: the mass percentage concentration is 25%-50%;

Alcohol reducing agent: The alcohol reducing agent is 2-mercaptoethanol or dithiothreitol, and the mass percentage concentration is 0.175%-0.35%;

8-Hydroxyquinoline: the mass percentage concentration is 0.05%-0.1%;

The pH of the dissociation solution is 7.0-8.5.

The combined application of the dissociation solution and the gel column has the following beneficial effects:

(1) Cells are lysed quickly, nucleic acid extraction can be completed with in 1 minute, and the nucleic acid extracted by the dissociation can be directly used in the PCR reaction, which greatly shortens the entire operation time of PCR nucleic acid diagnosis;

(2) Nucleic acid integrity can be maintained for a long time: from sample sampling, transportation to laboratory extraction of nucleic acid, it can maintain the integrity of nucleic acid in as little as three minutes and as long as one week;

(3) No need to be equipped with expensive equipment, just a handheld centrifuge.
